Tesofensine is a noradrenaline, dopamine, and serotonin reuptake inhibitor. This means that the medication prevents our synapses from taking these neurotransmitters away. In turn, we’re left with more noradrenaline, dopamine, and serotonin in our system.

This mechanism may sound familiar because mental health medications, including eating disorder medications, use a similar process. However, these medications don’t produce the weight loss many patients look for.

Originally, researchers looked at Tesofesine as a potential treatment for Parkinson’s and Alzheimer’s. However, in clinical trials, patients began losing too much weight. The extreme weight loss put the trial on hold.

Because of this finding, scientists began analyzing the medication for use in patients looking to lose weight.

How Tesofensine Works​

After shifting their focus toward Tesofensine as a weight loss drug, scientists found that it reduces the patient’s appetite. By stimulating our cholinergic system, tesofensine has become more successful than average weight loss remedies.

Our cholinergic systems regulate a variety of systems in our bodies, including sensory processing, attention, sleep, and arousal. So, stimulating this system allows our bodies to work faster than normal.

Additionally, Tesofensine increases the near transmission of monoaminergic neurotransmitters, which regulate energy balance. Each of the neurotransmitters it affects has a connection to obesity and depression. By increasing these neurotransmitters, Tesofenine is reducing the chance of both obesity and depression.

What Are the Tesofensine Benefits?​

There have been a few research studies that have looked at how Tesofensine works in a variety of patient cases. In these studies, we’ve learned a lot about Tesofensine:

  • It increases energy metabolism
  • It suppresses appetite
  • It improves patients’ quality of life

  • It leads to improved insulin sensitivity
  • It has anti-depressant effects

  • Sexual desire
Overall, the neurotransmitter controls our happiness, satisfaction, and optimism. Patients with too little serotonin are more likely to have conditions like chronic depression. They’re also more likely to overeat because they’re less likely to get satisfied quickly.

Tesofensine has a similar mechanism to SSRIs (selective serotonin reuptake inhibitors), which is the most common treatment for chronic depression. By preventing our bodies from picking up extra serotonin, these medications are supplying us with more free serotonin.

More serotonin increases our ability to feel happiness and satisfaction. So, we’re less likely to feel depressed and less likely to overeat.

Noradrenaline​

Noradrenaline, also known as norepinephrine, is both a neurotransmitter and a hormone. It plays a role in controlling the body’s flight-or-fight response.

Low levels of noradrenaline in the body can lead to sluggishness and fatigue. On the other hand, high levels of noradrenaline can increase fat loss and get the blood pumping faster.

By targeting this neurotransmitter, Tesofensine may be increasing patients’ metabolic rates. This means that patients can eat more without gaining extra weight.

At the same time, patients will be able to start burning fat more. Considering the dangerous effects of extra body fat, tesofensine can decrease risk factors associated with obesity. These include diabetes, cancer, hypertension, and insulin sensitivity.

Dopamine is a neurotransmitter that helps produce feelings of satisfaction and pleasure. When dopamine isn’t in the right balance, we can start losing the ability to feel satiated.

Those with diet-induced obesity may eat so much food that they begin dampening their dopamine receptors. This means that their bodies can no longer recognize whether they’re full or not, causing them to eat more than they normally would.

In fact, about 70% of patients who are obese have fewer dopamine receptors than patients of normal weight.

By inhibiting dopamine transport proteins (which get rid of dopamine), Tesofensine can increase the levels of dopamine in the brain. In turn, our bodies feel satisfied faster, leading to less eating.

Tesofensine may also be able to reverse dopamine blunting in patients with diet-induced obesity.


Dosage and Half Life and Interactions

The half life is 8-9 days so it does have a long half life

You may want to ease into stacking it with stimulants which could be amplified by using with tesofensine… It should stack quite well with nootropics, sarms, peptides and other fat burning compounds and supplements

Studies have shown a variety of effective doses… Generally they are over 6 month periods of time ranging from .25 mg per day once a day to 1 mg per day…the studies are showing that generally, over a 6 month period, there was up to 25 lbs lost… I see more .25 and .5 dosing used by most and seems to be tolerated well… There is still a lot of newer information coming so i would just be aware of your body and watch for unwanted side effects however from all i can conclude, this would be the best protocol to follow
